In the year 2075, Lela Dax visited the Caves of Mak'ala. (DS9 - The Lives of Dax short story: "First Steps")
In 2371, Benjamin Sisko and Julian Bashir brought Jadzia Dax to the Caves of Mak'ala, where Timor helped Jadzia to contact the Trill symbionts there. (DS9 episode: "Equilibrium")
In 2373, Ranul Keru began to work as a Guardian at the Caves of Mak'ala. (DS9 - Worlds of DS9 novel: Trill: Unjoined)
In 2375, Ezri Dax considered leaving Starfleet. Sisko advised her to begin caring for the symbionts in the Caves of Mak'ala. Dax ultimately remained in Starfleet. (DS9 episode: "Afterimage")

In 2406, President of Trill Otera Tul sent a communique to T'Vanik, Secretary General of the Federation Security Council, stating that the Trill government denied that any security breaches had occurred in the Caves of Mak'ala. The rumors were that the Undine (Species 8472) were using the caves to infiltrate the Alpha Quadrant from fluidic space. President Tul declined the offer of deploying Starfleet troops to the Caves of Mak'ala. (STO novel: The Needs of the Many)
In 3189, Adira, a Human who had been joined with a Trill symbiont named Tal, journeyed to the Caves of Mak'ala in order to enable them to access Tal's memories of former lives. Adira was accompanied by Commander Michael Burnham of the USS Discovery, who joined in Adira's communion with Tal's former hosts. These included Adira's late boyfriend, Gray Tal, and the Starfleet admiral Senna Tal. Adira emerged from the cave waters with Tal's memories restored, and became Adira Tal. The incident let to the acceptance of non-Trill hosts for Trill symbionts. (DSC episode: "Forget Me Not")
Mirror universe[]
Saavik entered the Caves of Mak'ala when Spock ordered her to investigate the Trill symbionts. When she found them infested by the Bluegill mutants, Spock ordered her to eradicate the symbionts. Agents of Imperial Starfleet subsequently hunted down every joined Trill in the galaxy, only leaving Curzon Dax alive. Before his death in 2293, Consul of the Terran Republic Spock recorded a confession. The litany of his crimes included the genocide on the Trill peoples. (TOS - Mirror Universe novel: The Sorrows of Empire)
Spock's confession was later played for the opening of the Galactic Commonwealth's parliament in 2378. (Mirror Universe novel: Rise Like Lions)
